Product Specific Information

The monoclonal antibody X2/45-12 reacts with human CD124 (IL-4R alpha chain).
CD124 can associate with the common gamma chain to form the type I IL-4R or with IL-13Rα1 to form the type II IL-4R.
Both receptor types bind IL-4, but only the type II receptor associates with IL-13.

• Type I IL-4 receptor: Expressed on T cells, B cells, NK cells, mast cells, basophils, and macrophages.
• Type II IL-4 receptor: Found on many non-hematopoietic cells and macrophages.

CD124 contains an ITIM that activates STAT6. IL-4 mediates high-affinity IgE antibody generation and Th2 polarization.
In mucosal barriers, IL-4 is involved in recruiting immune effector cells and inducing mucus overproduction.
A soluble form of IL-4R acts as a carrier protein to increase IL-4 half-life.

The X2/45 antibody has been reported to exhibit antagonist activity.

Applications Reported: Flow cytometry and Western blotting.
For functional assays, use the Functional Grade purified X2/45-12 (Product #16-1249-82).

Applications Tested:
This antibody has been tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage ≤1 µg per test (in 100 µL final volume). Optimal titration is advised for specific assays.

Target Information

IL4 Receptor alpha (IL4RA) is the alpha chain of the interleukin-4 receptor that binds IL-4 and IL-13, regulating IgE production, chemokine expression, and mucus production during allergic inflammation.
It promotes Th2 differentiation. The secreted extracellular domain (sIL4R alpha) inhibits IL-4-mediated proliferation and IL-5 upregulation but lacks signaling capability.
Genetic variations in IL4RA are associated with atopic conditions such as allergic rhinitis, sinusitis, asthma, and eczema.
