Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator. The 20S core consists of four rings of 28 non-identical subunits, with two rings of alpha and two of beta subunits. The 19S regulator includes a base (6 ATPase and 2 non-ATPase subunits) and a lid (up to 10 non-ATPase subunits). Proteasomes function in ATP/ubiquitin-dependent peptide cleavage in a non-lysosomal pathway.
A modified form, the immunoproteasome, processes class I MHC peptides and contains the 11S regulator (PA28) instead of the 19S regulator. The gamma subunit of the 11S regulator forms a homohexameric ring. Two transcript variants encoding different isoforms have been identified.
