The SH2D1A gene encodes a protein with a major role in bidirectional stimulation of T and B cells. It contains an SH2 domain and a short tail, associating with the signaling lymphocyte-activation molecule (SLAM) to inhibit recruitment of SHP-2. It also binds to surface molecules on activated T, B, and NK cells, modulating signal transduction. Mutations in this gene lead to X-linked lymphoproliferative syndrome type 1 (Duncan disease), a rare immunodeficiency associated with susceptibility to Epstein-Barr virus, severe mononucleosis, and malignant lymphoma. Multiple transcript variants encoding different isoforms are known.
