Product Specific Information

This antibody detects endogenous levels of IKK alpha/beta only when phosphorylated at Ser176/Ser177.

Target Information

IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) located on chromosomes 10 and 8, respectively, in humans. Upon stimulation by agents that activate the NF-κB pathway such as TNF alpha, the IKK complex subunits (IKK alpha, IKK beta) and regulator NEMO (IKK gamma) phosphorylate IκB proteins. This leads to ubiquitin-mediated degradation of IκB and phosphorylation of NF-κB, enabling its nuclear translocation. Phosphorylation at serine 176 and 180 in the activation loop induces a conformational change required for kinase activation.
