Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling and homing to lymphoid tissues.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry of mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Optimize empirically for best performance.

Fluorochrome Characteristics:
Super Bright 600 is excited by a 405 nm violet laser and emits at 600 nm. Use a 610/20 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal signal loss.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor composed of a C-type lectin domain, EGF-like domain, SCR sequences, a spacer, transmembrane, and cytoplasmic regions. It is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except some memory T cells.
