Target Information

Beta-catenin is an adherens junction (AJ) protein initially identified as a component of cell-cell adhesion structures. AJs are essential for forming and maintaining epithelial cell layers by regulating cell growth and adhesion between cells. Beta-catenin interacts with the cytoplasmic domain of E-cadherin, linking it to alpha-catenin, which anchors the complex to the actin cytoskeleton.

Studies show that beta-catenin also associates with cytoskeletal complexes containing adenomatous polyposis coli protein and microtubules, and participates in signaling pathways involving tyrosine kinases, phosphatases, and Wnt/Wingless. This interplay influences morphogenesis and cell proliferation.

Beta-catenin is expressed in various hair follicle cell types and epithelial cells, and its pathological roles have been identified in pilomatrixoma, medulloblastoma, colorectal and ovarian cancers, and other tumors. In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, stimulating genes such as cyclin D and c-myc that promote proliferation. This mechanism underlies its role in tumorigenesis of several solid tumors including breast, colon, liver, lung, gastric, prostate, and skin cancers.
