Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities essential for ER-based stress signaling.
IRE1 acts as a sensor for unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into an active transcriptional activator. It is critical for fetal viability and cell survival under stress conditions that cause protein misfolding in the ER.
Depletion of IRE1p has minimal effect under normal conditions but induces apoptosis under ER stress.
