Product Specific Information

The 4S.B3 monoclonal antibody reacts with human interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic properties. IFN-γ acts synergistically with TNF-α to inhibit cell proliferation and modulate immune responses across various cell types.

  • Applications Reported: Intracellular staining followed by flow cytometric analysis.
  • Applications Tested: Pre-diluted and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
    Recommended usage is 5 µL (0.25 µg) per test (100 µL final volume).
    Cell count: 10⁵–10⁸ cells/test (empirically determined).

Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is optimized for use on cytometers equipped with a 355 nm UV laser. Ensure instrument compatibility before use.

  • Buffer Recommendations: When using multiple polymer dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409) to minimize non-specific interactions.
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation: Compatible with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
  • Methanol Compatibility: Suitable for short-term methanol-based fixation (<1 hour).
  • Excitation/Emission: 355 nm / 732 nm; Laser: Ultraviolet.

Target Information

IFN-γ (Interferon gamma, Type II interferon) is a macrophage activation factor produced mainly by T-lymphocytes and NK cells. It is a 45 kDa homodimeric glycoprotein composed of two 146 amino acid subunits. IFN-γ exhibits species-specific activity and does not cross-react between human and mouse.

Functions include:

  • Antiviral and antiproliferative activity
  • Induction of MHC class I and II expression
  • Macrophage activation
  • Enhancement of immunoglobulin secretion by B cells
  • Cytokine regulation via JAK-STAT pathway activation

Human IFN-γ shows ~40% sequence homology with mouse IFN-γ. Mutations in the IFN-γ gene are associated with aplastic anemia.
