Product Specific Information

This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily containing an ITIM domain.
KLRG1 is expressed on subsets of γδ T cells, NK (CD56dim), CD8+ and CD4+ T cells, primarily in effector/memory phenotype cells that are short-lived but capable of immediate effector function.
Cadherin-E, -N, and -R are ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation. The percentage of KLRG1-positive cells can vary depending on antigen experience.

The clones 13F12F2 and 13A2 recognize similar epitopes based on cross-blocking studies.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended at 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm, optimized for UV (355 nm) laser-equipped cytometers. Ensure instrument compatibility.

Buffer Recommendations:
Use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) when combining multiple polymer dye-conjugated antibodies.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation Compatibility:
Samples can be fixed with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Compatible with short-term methanol-based fixation (<1 hour).

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric type II transmembrane receptor containing ITIM motifs that interact with SH2 domains of phosphatases such as SHP-1.
It acts as an inhibitory receptor expressed on NK cells and certain T cells, involved in tumor and virus-infected cell lysis and immune response regulation.
