Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 plays a critical role in peripheral tolerance and autoimmune disease in mice and is mainly expressed on activated T and B lymphocytes.
J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells and has been shown to modulate immune responses in vivo.
Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of stimulated mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Optimize antibody concentration for best performance.
Fluorophore Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is designed for UV (355 nm) laser-equipped cytometers. Protect from light due to photo-induced oxidation sensitivity.
Fixation Compatibility:
Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer or 1-step Fix/Lyse Solution. Compatible with short-term methanol-based fixation (≤1 hour).

Target Information
PD-1 (PDCD-1) is an inhibitory receptor expressed on activated T cells. It binds PD-L1 and PD-L2 on antigen-presenting cells to downregulate immune responses. In PD-1 deficient mice, hyperactivated immune systems and autoimmune diseases are observed.
Although PD-1 has a predicted molecular weight of 55 kDa, it may migrate at higher weight in SDS-PAGE.
