Target Information

The leucine-rich, glioma inactivated gene 4 (LGI4) belongs to the LGI family, which includes LGI1 as a representative member. This family comprises four highly related proteins containing leucine-rich repeats (LRRs) similar to transmembrane signaling molecules and receptors.
LGI1 functions as a candidate tumor suppressor gene for glioma and is associated with autosomal dominant lateral temporal epilepsy (ADTLE), characterized by focal seizures with auditory symptoms.
Although LGI4 shares high homology and expression patterns with LGI1, mutations in LGI4 have not been linked to ADTLE. The LGI4 gene resides in a region associated with benign familial infantile convulsions, and a GC-to-AT polymorphism has been correlated with childhood absence epilepsy.
Reduced LGI4 expression in cultured cells inhibits myelination, suggesting a role in neural development.
