Target Information
MCL1 (Myeloid cell leukemia-1)은 Bcl-2 패밀리에 속하며 세포 생존 유지, 분화, 프로그램된 세포사멸(apoptosis) 조절에 관여합니다.
Isoform 1은 apoptosis를 억제하고, Isoform 2는 이를 촉진합니다. MCL1의 C-말단은 bcl-2와 높은 서열 상동성을 가지며, DNA 손상 자극(방사선, 자외선, 알킬화제 등)에 의해 발현이 증가합니다.
MCL1은 미토콘드리아에 주로 존재하며, bcl-2와 유사하게 세포 생존을 촉진합니다.
Apoptosis 억제 기전은 명확히 밝혀지지 않았으나, Bim 또는 Bak과 같은 pro-apoptotic 단백질을 중화시키는 것으로 추정됩니다.
MCL1은 다양한 세포 유형에서 발현되며, 배아 발생 및 B/T 림프구 발달에 필수적입니다.
두 가지 주요 isoform(MCL1L, MCL1S)이 존재하며, MCL1S의 과발현은 세포 사멸을 촉진합니다.
