Product Specific Information

The OX39 monoclonal antibody reacts with mouse CD25 (IL-2 receptor alpha, 55 kDa). CD25 is expressed by early progenitors of T and B lineages and by activated mature T and B lymphocytes. It binds IL-2 with low affinity alone but forms a high-affinity receptor when associated with CD122 and CD132. CD25 plays a role in lymphocyte differentiation and activation/proliferation.

The OX39 clone has been shown to deplete CD25+ cells in vivo.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on rat splenocytes at ≤ 1 µg/test (100 µL final volume). Cell numbers may range from 10⁵–10⁸/test. Optimize titration for best performance.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Use a 710/50 bandpass filter. Ensure instrument compatibility.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD25 (IL2 receptor alpha chain / IL2RA):
A cytokine receptor involved in proliferation of T and B lymphocytes. Forms a high-affinity IL-2 receptor complex with IL2RB and IL2RG. Homodimeric IL2RA yields low-affinity receptor; IL2RB homodimer yields medium affinity. IL2 is produced primarily by mature T cells and acts as a growth and differentiation factor. Mutations in IL2RA are linked to immune dysregulation and colitis-like disease in mice.
