Product Specific Information

The HIT2 monoclonal antibody reacts with the human CD38 molecule, an approximately 45 kDa type II transmembrane protein expressed by thymocytes, peripheral B cells (including plasma cells), activated T cells, and monocytes. CD38 serves as a counter-receptor for CD31 and exhibits cyclase and hydrolase enzymatic activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test. One test = antibody amount for staining in 100 µL final volume, with 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser line (405 nm) and emits at 702 nm. Recommended detection filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light during handling and storage.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal effect on brightness and FRET efficiency.

Target Information

CD38 (NAD⁺ glycohydrolase) is a type II transmembrane glycoprotein that induces activation, proliferation, and differentiation of mature lymphocytes and mediates apoptosis of progenitor cells. It functions as an ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) and as a receptor.
Antibodies to CD38 are used for subtyping lymphomas/leukemias, plasma cell detection (e.g., myelomas), and as markers for activated B/T cells.
CD38 is highly expressed in the pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Associated diseases include chronic lymphocytic leukemia and Richter’s Syndrome.
