Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells throughout development except plasma cells. CD19 forms a complex with CD21, CD81, and MHC class II, contributing to BCR signaling, tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Internal testing indicates NovaFluor Yellow 570 may non-specifically stain B cells in SJL mice but not in BALB/c or C57BL/6 strains.

Applications Tested:
Validated on mouse splenocytes at ≤ 0.2 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Required for optimal staining. Add 5 µL per sample (10³–10⁸ cells). For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates are based on Phiton™ technology using engineered nucleic acid dye structures for optimized spectral performance with minimal spillover.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and some myeloid leukemia cells. CD19 acts as a co-receptor with BCR, regulating B cell activation and proliferation. Mutations cause hypogammaglobulinemia; overexpression leads to B cell hyperactivity.
