Product Specific Information

The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney podocytes, epithelial, and mesothelial cells. It is also a potent marker for lymphatic endothelium and thymic subcapsular epithelial cells. Podoplanin deficiency in mice causes respiratory defects and congenital lymphedema.

Suitable for flow cytometric analysis.

Applications Tested:
Validated using the TE-71 cell line. Recommended ≤0.125 µg per test (100 µL final volume). Optimal titration should be determined empirically.

Light Sensitivity:
The tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in:

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type glycoprotein (40–43 kDa) expressed in many tumors and normal cells, notably in lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal and thymic epithelial cells and plays a role in lymphatic endothelial regulation and podocyte morphology. Podoplanin interacts with CLEC-2 and correlates with tumor invasiveness. It contributes to cell migration, metastasis, and tumor invasion. Expression is regulated via Src activation of Cas and has been observed on Th17 cells and a subset of CD4+ PBMCs. Dysregulation is linked to lymphangioma and subglottic neoplasm and proposed as a marker for lung injury.
