Target Information

Botulinum neurotoxin type A (BoNT-A) is one of seven serotypes of Botulinum Neurotoxins produced by various strains of Clostridium botulinum.
BoNTs are synthesized as inactive single-chain precursors and activated by proteolytic cleavage into disulfide-linked two-chain proteins.

• Light Chain (50 kDa): Catalytic domain
• Heavy Chain (100 kDa): Translocation and receptor binding domains

BoNTs are the most potent protein toxins for humans. As zinc proteases, they cleave SNARE proteins, blocking acetylcholine release at the neuromuscular junction and causing flaccid paralysis.
E. coli-expressed recombinant light chains are active proteases but non-toxic due to lack of cell entry capability without heavy chains.
