Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 expression in a subset of B cells defines B-1 cells, a distinct lineage with unique developmental and functional properties. CD5 serves as a counter-receptor for CD72 and is involved in T-B cell interactions.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; optimal performance ≤0.25 µg/test (in 100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration is recommended for best results.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein found on mature T lymphocytes, most thymocytes, and many T-cell leukemias and lymphomas. It also reacts with a subpopulation of activated B cells and may regulate T-cell proliferation. CD5 is expressed on approximately 95% of thymocytes and 72% of peripheral blood lymphocytes, predominantly in T cell areas of lymph nodes. Associated diseases include thymus cancer and Richter’s Syndrome.
