Target Information

Thioredoxins (Trx) are small, multifunctional proteins with oxidoreductase activity, present in nearly all living cells. Trx contains a redox-active disulfide/dithiol group within the conserved Cys-Gly-Pro-Cys active site. The cysteine residues can form intramolecular disulfide bonds, which are reduced by Trx reductase using NADPH. Reduced Trx acts as a hydrogen donor for ribonucleotide reductase, essential for DNA synthesis, and functions as a general protein disulfide reductase. Specific reduction targets include protein disulfide-isomerase (PDI) and transcription factors such as p53, NF-kB, and AP-1. Trx also contributes to H₂O₂ removal when coupled with methionine sulfoxide reductase or peroxiredoxins.
