Product Specific Information

The monoclonal antibody H1.1 recognizes gp49, a receptor structurally similar to PIR and ILT/KIR receptor families.
Two closely linked genes on mouse chromosome 10 encode gp49A and gp49B, which share ~90% identity in extracellular regions but differ in cytoplasmic domains. gp49B contains an ITAM domain, while gp49A is short and lacks it.

gp49 expression was first identified on mast and LAK (IL-2 activated NK) cells, with LAK cells expressing predominantly gp49B. gp49 is also found on myeloid cells. Although its detailed function is not fully known, gp49B can inhibit cellular activation.

The H1.1 antibody recognizes both gp49A and gp49B. When crosslinked with anti-NK1.1, it can prevent cytokine release in KY-2 cells.
It has been reported for flow cytometric analysis and functional assays, and shown to block cytokine release.

Recommended usage:

  • Tested in flow cytometry using mouse bone marrow cells
  • ≤ 1 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)
  • Titrate for optimal performance

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4.
The encoded protein is a subfamily B LIR receptor, containing two or four extracellular Ig domains, a transmembrane domain, and ITIM motifs.
It is expressed on immune cells, binds MHC class I molecules, and transduces inhibitory signals that regulate immune activation.
It may also function in antigen capture and presentation, helping to control inflammation and prevent autoreactivity.
Multiple transcript variants encoding different isoforms have been identified (RefSeq, Jul 2008).
