Product Specific Information
The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38). CD244 is a 38 kDa protein expressed on NK cells, a subset of CD8+ T cells, γδ T cells, monocytes, basophils, and eosinophils. Binding of CD244 to its ligand CD48 results in NK cell activation and increased cytotoxicity and IFN-γ secretion.
Binding of the C1.7 monoclonal antibody and CD48 involves the same residue in the V domain of human CD244, explaining the activation of NK cells upon C1.7 binding. This interaction leads to tyrosine phosphorylation and recruitment of the adaptor molecule SAP (SLAM-associated protein). Patients with X-linked lymphoproliferative disorder (XLPD) have a mutation in SAP that prevents its binding to phosphorylated CD244.

Applications Tested: Flow cytometric analysis of normal human peripheral blood. Recommended ≤0.5 µg/test (100 µL final volume, 10^5–10^8 cells/test). Titrate for optimal performance.

Target Information
CD244 (2B4, SLAMF4) is a cell surface receptor on NK cells and some T cells that mediates non-MHC restricted killing. Interaction with SLAMF2 modulates NK-cell activity. CD244 dysfunction is associated with autoimmune diseases such as systemic lupus erythematosus and rheumatoid arthritis, and lymphoproliferative syndrome. Alternative splicing produces multiple isoforms.
