Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, associating with the BCR to regulate signaling, calcium flux, and proliferation of B cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes, ≤0.5 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions when multiple Super Bright dyes are used.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 2–8°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness.
Excitation/Emission: 405 nm / 600 nm

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and on certain myeloid leukemia cells. CD19 serves as a key regulator of B cell activation and proliferation, functioning as a signal-amplifying coreceptor for the B cell receptor (BCR). Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
