Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas.

The extracellular portion of CD22 contains seven Ig-like domains capable of binding sialic acid-containing ligands. The intracellular region includes tyrosine residues within ITIM and activation-like motifs that are phosphorylated upon B-cell receptor engagement, regulating B-cell receptor signaling.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells (5 µL/test). A test defines staining in 100 µL final volume for 10⁵–10⁸ cells/test.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) with emission at 600 nm. Recommended filter: 610/20 bandpass.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light-sensitive: protect vial and samples from light.
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed on B-lymphocytes. It appears early in B-cell differentiation, similar to CD19.
Membrane expression is limited to mature B cells (CD22+) and plasma cells (CD22−), useful for phenotyping mature leukemias. Strongly expressed in hairy cell leukemia and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.

CD22 preferentially binds α2,6-linked sialic acid, involved in regulation of B-cell receptor signaling through phosphorylation and recruitment of cytoplasmic phosphatases.
