Product Specific Information
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular biochemical pathways resulting in cellular activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse spleen cells at ≤0.25 µg/test (100 µL final volume). Optimal titration recommended.
Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). Recommended filter: 710/50 bandpass (alternative: 695/40). Ensure instrument compatibility.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. They associate with cytoplasmic signal transduction molecules via tyrosine-based motifs.
CD3 is present on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum. It is not expressed on B or NK cells. Reduced T lymphocyte percentages may be observed in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
