The eBioT21/8 monoclonal antibody reacts with the amino terminus of human CCR5 (CD195), an approximately 45 kDa G protein-coupled receptor (GPCR) that binds chemokines such as MIP-1α, MIP-1β, and RANTES. CCR5 is expressed on resting memory/effector T cells, monocytes, macrophages, and immature dendritic cells. It functions as a receptor directing lymphocyte chemotaxis and serves as the primary co-receptor for macrophage-tropic HIV-1, binding through gp120.

Applications reported include flow cytometry, immunoprecipitation, immunoblotting (WB), and immunocytochemical staining. It has also been used as a capture antibody in ELISA for detecting human CCR5 in cell lysates.

For flow cytometry, staining is recommended at 37°C for 1 hour in Flow Cytometry Staining Buffer (Product #00-4222-57). Use ≤0.5 µg per test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance. Binding may vary between donors and depend on staining conditions.

Target Information

CCR5 is a key co-receptor for HIV-1, HIV-2, and SIV infection, working alongside CD4 to mediate viral entry into host cells. It belongs to the chemokine receptor family (CCR5, CXCR4, CCR3, CCR2b, CCR8) and interacts with ligands such as RANTES, MIP-1α, and MIP-1β, which can inhibit M-tropic HIV-1 infection. CCR5 mRNA is expressed in lymphoid organs and monocytes, and its amino-terminal domain and extracellular loops serve as HIV binding sites.
