Product Specific Information
The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and is effective in treating renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for TCR assembly and surface expression. CD3 is expressed by thymocytes and all mature T cells.
Crosslinking of TCR initiates intracellular signaling leading to activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.5 µg)/test.

Laser: Violet Laser
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Use a 710/50 bandpass filter. Ensure instrument compatibility.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information
The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. They interact with cytoplasmic signal transduction molecules through tyrosine-based motifs.
CD3 antigen is found on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells. It is absent on B and NK cells. Reduced T cell percentages may occur in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
