Product Specific Information

This P4A10 monoclonal antibody reacts with canine CD25, the 55-kDa interleukin-2 receptor alpha (IL-2Rα) chain. In dogs, CD25 is expressed by early T and B lineage progenitors and activated mature lymphocytes. CD25 associates with IL-2Rβ (CD122) and IL-2Rγ (CD132) chains to form the high-affinity IL-2 receptor. Alone, CD25 binds IL-2 with low affinity and, along with CD4 and Foxp3, serves as a marker for regulatory T cells.

The P4A10 antibody recognizes the same epitope as the anti-human CD25 monoclonal antibody ACT-1, but with greater affinity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal canine peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the violet laser line (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD25 (IL2 receptor alpha chain / IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2RA forms part of a heterotrimeric complex with IL2RB and IL2RG, constituting the high-affinity IL-2 receptor. Homodimeric IL2RA forms low-affinity receptors, while IL2RB forms medium-affinity ones. IL2 expression in mature thymocytes is monoallelic, regulating precise gene expression. IL2 is mainly produced by mature T cells and functions as a growth, differentiation, and apoptosis regulator. It stimulates B-cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency.
