Target Information

IL-33 (Interleukin-33) is a 270-amino-acid protein belonging to the IL-1 family with an IL-1-like C-terminal domain. It functions both as a proinflammatory cytokine and as an intracellular nuclear factor with transcriptional regulatory properties. IL-33 binds to IL1RL1/ST2, recruiting MYD88, IRAK1, IRAK4, and TRAF6, and activates NF-kappaB and MAP kinases, promoting TH2-associated cytokine production. In vivo, IL-33 induces IL-4, IL-5, and IL-13 expression, leading to pathological changes in mucosal organs. It is highly expressed in high endothelial venules of tonsils, Peyer’s patches, and mesenteric lymph nodes, but nearly undetectable in placenta. IL-33 may co-localize with heterochromatin and act as a transcriptional repressor, showing dual functionality as both cytokine and nuclear factor. Despite its predicted molecular weight, IL-33 often runs at a higher molecular weight in SDS-PAGE.
