Product Specific Information

Description:
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5′-nucleotidase activity. Expression on myeloid cells (CD11b) is restricted to the bone marrow. In human cells, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling. Differences in CD73 expression patterns exist between mouse and human lymphocyte subsets.

CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. It is found on memory CD4 T cells and regulatory T cells, playing roles in immune regulation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes
• Recommended ≤ 0.125 µg/test (100 µL final volume)
• Optimal titration required for best performance

Important Usage Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover.

Note:
Internal testing shows that NovaFluor Yellow 690 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice.

Target Information

CD73 (Ecto-5′-nucleotidase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides (preferred substrate: AMP). It is a dimeric 70 kDa GPI-linked membrane protein used as a marker of lymphocyte differentiation.
Deficiencies in CD73 are associated with immunodeficiency diseases and tissue calcification disorders. The CD73 gene is localized to 6q14–q21 and has two transcript variants encoding different isoforms.
