Description:
The 1D11 monoclonal antibody reacts with human NKG2D, a 42 kDa lectin-like molecule expressed by NK cells, γδ T cells, CD8+ T cells, and some CD4+ T cells. NKG2D forms complexes with DAP10 and costimulates NK activation receptors. Its counter-receptors, MICA/MICB, are expressed on various epithelial tumors (lung, breast, kidney, ovary, prostate, colon carcinoma). Clones 5C6 and 1D11 block soluble MICA binding and inhibit lysis by γδ T cells.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL total volume). Recommended cell number: 10⁵–10⁸ cells/test.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)

• Excitation: 488–561 nm

Target Information

Natural killer (NK) cells mediate lysis of tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express calcium-dependent (C-type) lectins involved in NK function regulation. The NKG2 gene family encodes type II transmembrane proteins with C-type lectin domains and is located within the NK complex region.
