Description:
Monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, mesenchymal stromal cells, and certain tumors such as melanoma. Expression on circulating blood cells is limited to a subset of T cells. The protein is heavily glycosylated, with more than 50% of its mass from carbohydrates. P1H12 cross-reacts with mouse, rabbit, and dog, but not rat.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Tested:

• Pre-titrated and tested by flow cytometry with normal human peripheral blood cells and HUVEC
• Recommended use: 5 µL (0.02 µg) per test (100 µL total volume)
• Suitable for 10⁵–10⁸ cells/test

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.

Target Information

CD146 (MCAM):
An integral membrane glycoprotein of the immunoglobulin superfamily found at cell-cell junctions in endothelial cells and other cell types. CD146 functions as a calcium-independent adhesion molecule and may act as a signal transduction molecule via Fyn kinase activation, influencing actin cytoskeleton assembly. It contributes to endothelial cohesion and may facilitate tumor metastasis by mediating interactions with the vascular system. CD146 also plays roles in neural crest cell adhesion during embryonic development and intracellular calcium signaling.
