Product Specific Information

The monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, and some tumors (especially melanoma). It is also found on mesenchymal and endometrial stromal cells, and a subset of T cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells and HUVEC. Recommended use: 5 µL (0.02 µg) per test in 100 µL final volume.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before mixing with cells.

NovaFluor Conjugate Info:

• Based on Phiton™ technology, utilizing nucleic acid dye structures engineered for minimized spillover and spread.
• Excitation: 509 nm
• Emission: 665 nm

Target Information

CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal and malignant cell types.
Functions include:

• Calcium-independent cell adhesion
• Signal transduction via Fyn kinase recruitment
• Regulation of actin cytoskeleton assembly
• Maintenance of endothelial monolayer cohesion
• Facilitating melanoma cell interaction with vascular elements for tumor spread
• Potential role in neural crest cell adhesion during embryonic development
• Surface receptor triggering tyrosine phosphorylation of FYN and PTK2
