Description:
Monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, and Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, and some tumors (notably melanoma). It is also expressed on mesenchymal and endometrial stromal cells. Expression on circulating cells is limited to a subset of T cells, not endothelial cells, as CD31 and CD51/61 are negative. The protein is heavily glycosylated, with over 50% of its mass derived from carbohydrates. P1H12 cross-reacts with mouse, rabbit, and canine, but not rat.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated using normal human peripheral blood cells and HUVEC. Recommended usage is 5 µL (0.02 µg) per 100 µL test volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining.
• Add 5 µL of CellBlox Plus per sample to antibody cocktails prior to combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD146 (MCAM):
An integral membrane glycoprotein of the immunoglobulin superfamily, expressed at cell-cell junctions in endothelial cells and various normal and malignant cell types. CD146 functions as a calcium-independent adhesion molecule and participates in signal transduction via Fyn kinase, influencing actin cytoskeleton assembly. It plays key roles in endothelial cohesion, tumor metastasis (especially melanoma), and neural crest cell adhesion during development. It also mediates tyrosine phosphorylation of FYN and PTK2 and transient calcium signaling.
