Target Information
iNOS (Inducible Nitric Oxide Synthase) is an enzyme responsible for the production of nitric oxide (NO), a gaseous free radical that mediates various cellular signals such as vasorelaxation, neurotransmission, and cytotoxicity.
iNOS expression is inducible in liver and other tissues by lipopolysaccharide and cytokines. It catalyzes the oxidation of L-arginine to produce L-citrulline and NO.
There are three NOS isoforms:
- nNOS (type I) – neuronal
- eNOS (type III) – endothelial
- iNOS (type II) – inducible
All isoforms contain calmodulin, NADPH, FAD, and FMN binding domains. iNOS is expressed in macrophages, hepatocytes, synoviocytes, and smooth muscle cells. Cytokines such as IFN-γ, TNF, IL-1, IL-2, and LPS increase iNOS mRNA and protein levels. Protein kinase C activators also enhance iNOS activity.
After cytokine induction, iNOS shows delayed activation followed by sustained NO production.
Three iNOS pseudogenes are found within the Smith-Magenis syndrome region on chromosome 17.
Dysfunction of iNOS is associated with diseases such as achalasia and impotence.
