Product Specific Information

The SB/199 monoclonal antibody reacts with mouse CD127, the high-affinity α subunit of the mouse IL-7 receptor.
IL-7 receptor α chain is expressed by immature B cells in bone marrow, double-negative (CD4-/CD8-), single-positive (CD4+ and CD8+), but not double-positive (CD4+/CD8+) thymocytes. Mature peripheral T cells express low levels of CD127. SB/199 binding blocks A7R34 binding, another antibody recognizing mouse CD127.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤0.25 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimal titration recommended.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Can replace PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (alternative: 695/40).

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be empirically determined.

Target Information

CD127 (Interleukin-7 receptor, IL-7R) is a glycoprotein regulating lymphopoiesis. The active receptor is an α/γ chain heterodimer.

• α chain: Determines specific signaling events through cytoplasmic molecule association.
• γ chain: Activates signal transduction; also associates with IL-2 receptor.
  CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
  Defects in CD127 are linked to severe combined immunodeficiency (SCID) variants.
