Members of the STE-20 like kinase family are known to stimulate MAPK pathways by directly activating MAPKKK. STRAD is a pseudokinase with a STE-20-like domain but lacks several residues required for catalytic activity. It binds specifically to LKB1 and regulates its tumor suppressor functions. STRAD acts as an upstream activator and directs the subcellular localization of LKB1 by anchoring it in the cytoplasm. The STRAD-LKB1 interaction results in phosphorylation of STRAD and enhanced autophosphorylation of LKB1.
