Product Specific Information

Description:
The eB121-15F9 monoclonal antibody reacts with the mu heavy chain of mouse IgM. It does not react with other classes of mouse immunoglobulin including IgD, IgG, or IgA. IgM is expressed intracellularly during early stages of B lymphopoiesis and on the surface of mature B cells in bone marrow and peripheral tissues. Fluorochrome-conjugated eB121-15F9 can be used as a detection secondary for mouse IgM.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells. Recommended use ≤ 0.5 µg/test (in 100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test and should be empirically determined. Optimal performance requires titration.

Fluorochrome Information:
Super Bright 436 can be excited with a 405 nm violet laser and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

Mouse IgM is an immunoglobulin M antibody produced by B cells in response to antigens. It is the first antibody produced during an immune response and is the largest immunoglobulin in serum. Composed of five heavy and five light chains, IgM activates complement and agglutinates antigens. It plays a key role in the innate immune response and is widely used in research applications such as ELISA, Western blotting, and immunohistochemistry.
