Product Specific Information
The monoclonal antibody 3D10 recognizes Ly-49H, a member of the C-type lectin Ly-49 multigene family of receptors found on natural killer (NK) cells.
Ly-49H, like Ly-49D, lacks the ITIM (immunoreceptor tyrosine-based inhibitory motif) and contains an arginine in the transmembrane domain, indicating its role as an activation receptor.
Ly-49H is expressed in a subset of NK cells but not in NKT (NK1.1⁺CD3⁺) cells. NK cells can express several Ly-49 proteins.
Preferential staining of Ly-49H with Ly-49D has been observed (Product #00-5523-00).
For staining protocol, refer to BestProtocols® – Staining intracellular antigens for flow cytometry (Protocol B).
- Recommended usage: ≤ 0.125 µg/test
- Test volume: 100 µL
- Cell count: 10⁵–10⁸ cells/test (empirically determined)
- Optimize antibody titration for best performance
- Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
Ly49H is a C-type lectin receptor family member found on NK cells.
Like Ly49D, it lacks ITAM and contains an arginine residue in the transmembrane domain, functioning as an activating receptor.
Expressed in NK cell subsets, not in NKT (NK1.1/CD3⁺) cells.
NK cells may express multiple Ly49 receptors.
