Target Information
CtIP is a 125 kDa protein originally identified through its interaction with the transcriptional repressor CtBP via the PLDLS motif, suggesting a role in transcription regulation. It also interacts with BRCA1 through the C-terminal BRCT domains, indicating potential tumor suppressor functions. The CtIP–BRCA1 interaction can be disrupted by DNA-damaging agents such as UV or gamma irradiation. Li et al. (Nature 406, 210–215, 2000) demonstrated that ATM phosphorylates CtIP at serine residues 664 and 745; mutation at these sites prevents BRCA1 dissociation from CtIP, leading to persistent repression of BRCA1-dependent induction of GADD45 following ionizing radiation.
