Target Information

CtIP is a 125 kDa protein originally identified as interacting with the transcription repressor CtBP via the PLDLS motif, suggesting a role in transcription regulation. It also interacts with BRCA1 through the C-terminal BRCT domains, implying a potential tumor suppressor function. The CtIP–BRCA1 interaction can be disrupted by DNA damage such as UV or gamma irradiation. Studies (Li et al., Nature 406, 210–215, 2000) demonstrated that ATM phosphorylates CtIP at serine residues 664 and 745; mutation at these sites prevents BRCA1 dissociation from CtIP, leading to persistent repression of BRCA1-dependent GADD45 induction upon ionizing radiation.
