Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a critical role in T cell development and activation. RM4-5 binding is blocked by GK1.5.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Flow cytometric analysis of mouse splenocytes (≤ 1.0 µg/test)
    • A test is defined as staining a cell sample in 100 µL final volume.
    • Cell number range: 10⁵–10⁸ cells/test.
    • Optimize titration for best performance.

Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is designed for cytometers with a 355 nm UV laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

  • Super Bright Complete Staining Buffer (Product # SB-4401-42)

Light Sensitivity: Protect vial and stained samples from light due to photo-induced oxidation.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed on T helper cells, and also found on monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is detected in thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. CD4 aids early T-cell activation through association with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in infectious and immune CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
