Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.
- This antibody shows broad species reactivity.
Target Information
PGP9.5 (Protein gene product 9.5, also known as UCH-L1 or PARK5) is a neuron-specific protein, distinct from neuron-specific enolase. It has a molecular weight of 27 kDa and belongs to the ubiquitin carboxyl-terminal hydrolase family 1 (peptidase family C12).
PGP9.5 exhibits both ubiquitin hydrolase and ligase activities, hydrolyzing small C-terminal adducts of ubiquitin to generate monomers. It is widely present in neurons, nerve fibers, neuroendocrine cells, renal tubules, spermatogonia, Leydig cells, and ovarian tissues.
Altered expression of PGP9.5 is associated with several diseases: overexpression with non-small cell lung cancer and decreased expression with Huntington and Alzheimer diseases.
It serves as a useful neuronal marker in studies of neurodegenerative disorders such as Parkinson’s disease.
