Product Specific Information
The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and has been proven effective in the treatment of renal, heart, and liver allograft rejection.
CD3 subunits (gamma, delta, epsilon) are essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Crosslinking of TCR initiates intracellular signaling leading to activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells at ≤ 0.06 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.
Target Information
CD3 subunit complex is critical for transducing antigen-recognition signals and regulating TCR surface expression. The CD3 gamma, delta, epsilon, and zeta subunits are structurally related immunoglobulin superfamily members encoded on chromosome 11.
CD3 components associate with intracellular signaling molecules via tyrosine-based motifs. CD3 is expressed on most T cells (68–82% of lymphocytes, 65–85% of thymocytes) and Purkinje cells but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
