Product Specific Information
EpCAM (Epithelial cell adhesion molecule, CD326, KSA, TROP1) is a 40 kDa cell-surface adhesion molecule involved in homophilic, calcium-independent cell-cell interactions. It is a type-I transmembrane protein expressed mainly on the basolateral surface of most epithelia.
Normal epithelia express low levels of EpCAM, but increased expression correlates with enhanced proliferation and transition to a mesenchymal phenotype. EpCAM serves as a diagnostic marker for circulating metastatic carcinoma cells, while non-epithelial cancer cells do not express EpCAM.
Applications Reported:
This 1B7 antibody is reported for use in flow cytometric analysis and immunohistochemical staining of frozen tissue sections.
Applications Tested:
- Flow cytometric analysis of A549 cell line: ≤ 0.25 µg/test (100 µL final volume; 10⁵–10⁸ cells/test).
- Immunohistochemistry of frozen tissue: ≤ 10 µg/mL.
Antibody should be titrated for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Ep-CAM (epithelial adhesion molecule, epithelial specific antigen, ESA) is a transmembrane glycoprotein (~40 kDa) expressed in epithelial tissues. It mediates Ca²⁺-independent homotypic interactions and influences cell cycle, proliferation, and metabolism through induction of proto-oncogene c-myc and cyclin A/E.
Ep-CAM-mediated adhesions negatively regulate cadherin-mediated adhesions, affecting epithelial differentiation and growth. Overexpression is associated with enhanced epithelial proliferation and is a marker for epithelial tumors. Ep-CAM is expressed on basolateral surfaces in most simple epithelia and carcinoma types, and distinguishes adenocarcinomas from pleural mesotheliomas.
