Product Specific Information
This TM-724 monoclonal antibody reacts with human Autoimmune Regulator (AIRE), a 58 kDa transcription factor highly expressed within the perinuclear region of mature medullary thymic epithelial cells (mTECs).
AIRE mRNA has been observed in lymph node, fetal liver, and intestines, as well as in dendritic cells, epithelial cells, and PBMCs.
AIRE is involved in thymocyte development and regulation of tissue-specific antigen expression during negative selection, as well as chemokine expression and antigen presentation.
Three isoforms (AIRE-1, AIRE-2, AIRE-3) have been reported, all predicted to be recognized by TM-724.
Mutations in AIRE are associated with autoimmune-polyendocrinopathy-candidiasis-ectodermal-dystrophy (APECED) and autoimmune polyglandular syndrome type 1 (APS-1).

Target Information
AIRE is a transcriptional regulator protein that binds to DNA as a dimer and tetramer, not as a monomer, and regulates gene expression.
It is a nuclear 545 amino acid protein with HSR domain, two PHD-type zinc fingers, and a SAND domain, forming high-molecular-weight complexes.
AIRE interacts with transcriptional coactivators CBP and CREBBP, forms homodimers/tetramers, and nuclear bodies.
Three isoforms are reported, one predicted to undergo nuclear-mediated decay (NMD).
AIRE mediates E3 ubiquitin ligase activity and defects cause the rare autosomal-recessive autoimmune disease APECED.
