Product Specific Information

Description:
The RMST2-2 monoclonal antibody reacts with ST2 (IL-33 receptor, IL-33R, or interleukin 1 receptor-like 1), a member of the IL-1 receptor family. The membrane-bound form has three extracellular immunoglobulin domains and an intracellular toll-interleukin-1 receptor domain. ST2 dimerizes with IL-1R accessory protein in a ligand-dependent manner, and IL-33/ST2 interaction induces TH2 cytokine production. The soluble isoform acts as a decoy receptor, blocking IL-33/ST2 signaling.

ST2 is expressed by TH2 lymphocytes, mast cells, eosinophils, basophils, innate lymphocytes, smooth muscle, and endothelial cells, and is involved in host defense, allergy, and inflammation. The IL-33/ST2 interaction has been shown to be atheroprotective.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow-derived mast cells. Recommended ≤0.5 µg per test (in 100 µL final volume, cell number 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.


Target Information

IL-33R (ST2):
Receptor for IL-33 and member of the IL-1 receptor family. Its extracellular domain contains three Ig-like domains that interact with IL-1RAcP and IL-33. IL-33 ligation activates MyD88, IRAK1, IRAK4, and TRAF6, leading to NF-κB and MAPK pathway activation and release of cytokines such as IL-5, IL-6, IL-8, and IL-13.
Two main isoforms exist: ST2L (membrane-bound) and ST2S (soluble). The soluble form acts as a decoy receptor, dampening IL-33 signaling. Expressed mainly by mast cells, basophils, eosinophils, and in mice also by ILC2 and Th2 cells. IL-33R signaling is implicated in allergies, arthritis, atherosclerosis, and sepsis.
