Target Information

Cadherins are a multigene family of Ca++-dependent cell adhesion molecules. They are transmembrane glycoproteins consisting of an extracellular domain, which mediates Ca++-dependent intercellular adhesion by homophilic interactions, a transmembrane region, and a cytoplasmic domain. The extracellular domain is divided into subdomains EC1–EC5. Homologies among cadherins are most prominent in the cytoplasmic domain and EC1–EC2 regions, and less so in EC5 or the transmembrane region. The adhesive function is primarily determined by the N-terminal portion of the molecule.

Four major cadherins—neuronal (N), epithelial (E), placental (P), and muscle (M)—have been identified in mammalian cells. M-cadherin is not expressed in fibroblasts but is present at low levels in myoblasts and is upregulated during myotube formation, indicating its role in skeletal muscle cell differentiation.
