Target Information

Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. This family cleaves most prohormones and neuropeptide precursors, as well as various cellular, viral, and bacterial proteins transported via the constitutive secretory pathway.

Furin and other PCs share structural similarities:

  • ~10 kDa amino-terminal proregion
  • ~55 kDa conserved subtilisin-like catalytic domain
  • Variable carboxyl-terminal domain

These enzymes become active after proregion cleavage within the correct cellular compartment. Furin is unique among PCs in possessing a transmembrane domain. Cleavage occurs at the carboxyl-terminus of the consensus sequence RX(K/R)R.

The acidic peptide sequence C771PSDSEEDEG780 localizes furin to the trans-Golgi network, and phosphorylation of serine residues in this region modulates intracellular routing. The tetrapeptide sequence Y759KGL762 directs internalization from the cell surface.
