Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and plays an essential role in brain development. Mutations, rearrangements, and amplifications in the ALK gene have been associated with various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement often leads to the formation of a fusion gene combining ALK and nucleophosmin (NPM), where the 3′ region of ALK (chromosome 2) fuses with the 5′ region of NPM (chromosome 5). The resulting NPM-ALK fusion protein is oncogenic.
ALK encodes a receptor protein-tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK fusion product. ALK shows high sequence similarity to LTK and exerts its effects on specific neurons during brain development.
