Target Information

G protein-coupled receptor GPR49 (LGR5) is expressed in brain, skeletal muscle, placenta, and spinal cord. ESTs have been isolated from normal human brain (amygdala), embryo, and placenta libraries, as well as from germ cell, uterus, and brain libraries.
G-protein Coupled Receptors (GPCRs) are one of the largest families of signaling molecules, with more than a thousand members predicted. They share a structural motif of seven membrane-spanning helices and exist in both active and inactive forms.
Various ligands activate GPCRs, leading to signaling via G-proteins and downstream effectors. Studies also describe mechanisms of GPCR negative regulation, which are important for cellular physiology and pharmacological research.
Notably, GPCR-targeted drugs such as Zyprexa (Eli Lilly) and Claritin (Schering-Plough) have achieved significant success in mental health and allergy therapeutics.
